Non-invasive online monitoring of biomarkers of particulate toxicity in China

Recently, Prof. Mao Sheng from the School of Environmental Science and Engineering of Peking University and Prof. Zheng Yufeng from the Department of Chemistry of Fudan University collaborated to create a dLABer system using live rat exposed exhaled gas sampling, microfluidic and commercial silicon nanowire biosensors to realize real-time monitoring of living organisms. Exposure of biomarkers to exhaled air pollutants can be directly used for non-invasive online research on the health effects of air pollution.

In order to verify the system, the research team first injected the same amount of atmospheric particulate matter from different sources, and then used the dLABer system to monitor the level of inflammatory factor interleukin-6 (IL-6) in the exhaled breath of rats. The study found that the level of IL-6 produced by exposure to particulate matter from different sources was up to 1000-fold different, which was consistent with the trend of analyzing IL-6 levels in rat blood samples by enzyme-linked immunosorbent assay (ELISA). At the same time, it was also found that the results of PM toxicity obtained by different methods such as dithiothreitol (DTT) were consistent with those obtained by dLABer. The dLABer system developed by the research group has several orders of magnitude higher signal-to-noise ratio than ELISA, and can continuously monitor biomarker levels exhaled in living animals. The real-time monitoring of biomarkers in exhaled breath by dLABer not only helps us to understand the damage of air pollutants to the respiratory system (respiratory tract and lungs), but also provides a powerful basis for understanding the pathophysiological processes related to the respiratory system.
