pharmacological action
Pemetrexed enters the cells through folate carriers and the folate binding protein transport system on the cell membrane. Once pemetrexed enters the cell, it transforms into a form of glutamic acid under the action of the leaf glutamic acid synthetase. Polyglutamate is retained in cells as an inhibitor of thymidylate synthase and glycyamine nucleotidyl transferase. Glutamic acid showed a time - concentration dependent process in tumor cells, but the concentration in normal tissues was very low. The longer the half-life of multiple glutamic metabolites in tumor cells, the longer the time of action of drugs in tumor cells.
Preclinical studies showed that pemetrexed could inhibit the growth of mesothelioma cell lines (MSTO? 211H, NCI? H2052) in vitro. Mesothelioma cell line MSTO? 211H has shown synergistic effect of pemetrexed combined with cisplatin.
